چکیده

Enzyme-induced carbonate precipitation (EICP) is a new biogeotechnical ground improvement technique that uses calcium (CaCO3) formed by biochemical processes to increase soil strength and stiffness. In this paper, crude urease extracted from soybeans was employed catalyze the of CaCO3 in sand. To optimize extraction efficiency, factors affecting soybean extraction, including powdered particle size, concentration, soaking time, temperature, were addressed. This paper also provided further insight regarding impact activity extract on chemical conversion efficiency biocementation performance EICP. The findings revealed concentration size two most important extract. enzyme utilized EICP process might lead different reactant efficiencies urea-CaCl2 solution, consequently, physical mechanical properties biocemented Considering performance, 60 g/L concluded as preferred quantity for extracting urease, with an 6.62 mM urea min−1. Under condition, approximately 95% 0.5 M urea-0.5 CaCl2 could be obtained merely 12 h, unconfined compressive (UCS) EICP-treated sand exceeded 4 MPa content ~8%. As high-efficient cost-effective alternative purified precipitation, showed great potential improvement.

An Overview of the Factors Affecting Microbial-Induced Calcite Precipitation and its Potential Application in Soil Improvement

Microbial-induced calcite precipitation (MICP) is a relatively green and sustainable soil improvement technique. It utilizes biochemical process that exists naturally in soil to improve engineering properties of soils. The calcite precipitation process is uplifted by the mean of injecting higher concentration of urease positive bacteria and reagents into the soil. The main objective of this pap...

Soybean leaf urease: a seed enzyme?

The soybean (Glycine max L. [Merrill]) var Itachi has 0.2 to 0.3% the urease activity found in developing embryos of a normal line, Prize. The hydroxyurea sensitivity and pH preference of this basal seed urease indicate that it represents a unique enzyme rather than an unusually low level of the normal seed urease. Itachi's seed urease is less sensitive to hydroxyurea inhibition (65-80% inhibit...

Urease activity in microbiologically-induced calcite precipitation.

The role of microbial urease in calcite precipitation was studied utilizing a recombinant Escherichia coli HB101 containing a plasmid, pBU11, that encodes Bacillus pasteurii urease. The calcite precipitation by E. coli HB101 (pBU11) was significant although its precipitation level was not as high as that by B. pasteurii. Addition of low concentrations (5-100 microM) of nickel, the cofactor of u...

Halotolerant, alkaliphilic urease-producing bacteria from different climate zones and their application for biocementation of sand.

Microbially induced calcium carbonate precipitation (MICP) is a phenomenon based on urease activity of halotolerant and alkaliphilic microorganisms that can be used for the soil bioclogging and biocementation in geotechnical engineering. However, enrichment cultures produced from indigenous soil bacteria cannot be used for large-scale MICP because their urease activity decreased with the rate a...
